دزسنجی زیستی با استفاده از سنجش ریزهسته

چکیده

پیشرفت سریع و روز افزون کاربرد پرتوها در زمینه­های مختلف پزشکی، صنعتی و افزایش تأسیسات و فعالیت­های هسته­ای، افراد شاغل در این مراکز را در معرض خطر بالاتر پرتوگیری نسبت به گذشته قرار می­دهد. با توجه به خطرات زیستی و تأثیرات سوء این پرتوگیری­ها بر سلامت افراد، تخمین دز واقعی رسیده به بدن برای اتخاذ تدابیر درمانی مناسب و به­موقع، حائز اهمیت فراوان است. در حوادث پرتویی با ابعاد وسیع که نیاز به دسته­بندی سریع مصدومین است، استفاده از روش سنجش ریزهسته و دزسنجی زیستی بر اساس تهیه منحنی دز- پاسخ بسیار ارزشمند بوده و از کارایی بالایی برخوردار است. برای تهیه و رسم منحنی دز- پاسخ با این روش، نمونه خون از 16 نفر زن و مرد سالم در دو گروه سنی مختلف گرفته شد و تحت تابش پرتو گاما با دزهای 0، 25/0، 5/0، 1، 2 و 4 گری قرار گرفتند. پس از کشت و محصول برداری و تهیه لام، تعداد ریزهسته­ها در هر دز شمارش و منحنی دز- پاسخ رسم شد که از تابع خطی درجه دو تبعیت می­کرد. جهت بررسی کارآیی منحنی به­دست آمده، تخمین دز در دو مورد حادثه پرتوگیری شغلی و دو نمونه خون پرتودهی شده با دزهای نامشخص با هر دو روش استاندارد بررسی متافاز (شکست های کروموزومی دو سانترومری) و ریزهسته انجام شد و نتایج آن مورد مقایسه قرار گرفت. تطابق بسیار خوب نتایج حاصل از هر دو روش، نشان­دهنده کارایی بالای روش سنجش ریزهسته در تخمین دز زیستی است.

Biological dose estimation of ionizing radiation by using micronucleus assay

چکیده [English]

Rapid development in nuclear industry and medical applications involving radiation sources would increase the potential of exposure to the ionizing radiation. Due to the harmful effects of radiation exposure on the health of radiation workers, rapid dose estimation of the overexposed victims with high accuracy is quite important in taking appropriate medical treatment plan. Biological dosimetry based on micronucleous (MN) assay is a perfect technique with unique advantages for a fast triage in radiation accidents. To develop the dose response curve, blood samples of 16 healthy individuals in two groups of male and female and in two age ranges were collected and exposed to 0, 0.25, 0.5, 1, 2 and 4 Gy gamma rays, cultured, harvested, slides prepared and micronuclei in each dose scored. The results showed that the curve follow linear quadratic formula. In order to validate the performance of created dose response curve, two real accidental over-exposed workers and two unknown irradiated blood samples were investigated by newly established curve and compared with results obtained by metaphase analysis (dicentric). The doses estimated by MN assay showed a very good conformity with those estimated by dicentric analysis, pointing out that the MN assay is a reliable, easier and valuable alternative method for biological dosimetry.
