Radio protective effects of Cerium Salt against Gamma-ray-induced structural changes in Bovine Serum Albumin

Abstract

Ionizing radiation such as γ-radiation causes deleterious effects on protein.In this research,the effect of gamma radiation in the therapeutic dose of3 Gy and radioprotection of cerium salt (H8N8CeO18) on the structure and surface charge of bovine serum albumin (BSA) was studied. The first, secondary, tertiary structure and surface charge of BSA using UV-Vis spectroscopy, Circular dichroism (CD) and electrophoretic light scattering (ELS) were investigated. The results show that covalent bounds of protein were not destroyed in dose of 3 Gy of gamma irradiation indicating that the first structure of BSA is preserved. But, the secondary and tertiary structures of irradiated BSA proteins has been changed. The result of CD studies shows a 7% decrease in α-helix and an increase in other secondary structures for irradiated BSA in comparison to non- irradiated ones. Results also show that Cerium salt can be a strong radio protector to inhibit changes of secondary and tertiary structures of irradiated BSA protein.
